Abstract:

The datasets contain larval size, water temperature, cell cycle data, and classification of larval condition.

Purpose:

The datasets were developed to assess the condition of Walleye Pollock larvae using larval size, water temperature, and cell cycle data measured with flow cytometry.

The datasets contain larval standard length measured in mm, the proportion of cells in the S and G2/M phases of the cell cycle, and the ratio of the number S phase nuclei to the number of G1 phase nuclei with high nRNA content measured with flow cytometry. Also included is the mean water temperature (°C) between the surface and 40 m depth.

Larvae were collected using a 60 cm bongo frame fitted with 0.505 mm mesh nets and were frozen at -80°C. Tissue preparation, flow cytometry protocol, and data analysis are described in Porter and Bailey (2011 and 2013).

A BD Biosciences Influx flow cytometer (BD Biosciences, San Jose, CA) was used to analyze each larva. The proportion of nuclei in the S, and G2/M phases was calculated for each larva with MultiCycle AV software, vers. 4.0 (Phoenix Flow Systems, San Diego, CA). FCS Express flow cytometry analysis software, vers. 3.0 (De Novo Software, Los Angeles, CA) was used to calculate the ratio of the number S phase nuclei to the number of G1 phase nuclei with high nRNA content. A detailed description of the analyses is given in Porter and Bailey 2013.
